Patients: This Phase I study involved 53 patients with glioblastoma multiforme, 16 patients with anaplastic astrocytoma, 3 patients with anaplastic oligodendroglioma, and 1 patient with an anaplastic mixed glioma.

Treatment: Treatment consisted of two chemotherapy drugs, irinotecan plus BCNU.

Toxicity: There were five deaths associated with this treatment: pulmonary fibrosis confirmed by autopsy, pulmonary embolism, cardiopulmonary arrest, cerebral herniation, and infection. The authors concluded that three of the five deaths were “possibly drug related.” Other toxicities included: decreased DLCO, infiltrates, diarrhea, nausea/vomiting, vasovagal episode, infection with neutropenia, and hematologic toxicity.

Results: There were no complete responses and overall survival was not discussed.
