Patients: This Phase I study involved 32 patients with advanced solid-tissue tumors, and 31 were evaluated. Twenty-three of these patients were men, and nine were women. The median age was 58 years. Twenty-two patients (69%) had colorectal cancer (asymptomatic or with mild symptoms).

Treatment: The treatment consisted of chemotherapeutic drug OSI-7904L This drug blocks an enzyme (thymidylate synthase) involved in DNA synthesis. The drug is contained inside liposomes to localize the drug to tumor cells more efficiently.

Toxicities: At the highest dosage given, one patient died after having the following stage 3-4 toxicities: stomatitis, fatigue, tachyarrhythmia, rash, hand-foot syndrome, diarrhea, and neutropenia. For the next-highest dosage given in this study (given to 10 patients), grade 1-3 toxicities were anemia, thrombocytopenia, dyspnea, fatigue, and skin rashes.

Results: There were no objective responses.
