Patients: This Phase I study involved 20 patients with previously untreated glioblastoma multiforme and one patient with a rhabdoid tumor.

Treatment: Treatment consisted of surgery or stereotactic biopsy followed by cranial radiotherapy and the chemotherapy drug, topotecan (hycamtin).

Toxicity: Toxicities included neutropenia (grade 3-4), leucopenia (grade 3), anemia (grade 3), thrombocytopenia (grade 3), and vomiting (grade 3).

Results: Median survival for all patients (from date of initial surgery) was 69 weeks (16.0 months).

Support: GlaxoSmithKline provided topotecan and logistical assistance, and Biotrial contributed to the “realization of the study.” GlaxoSmithKline markets topotecan.
