Description:
Patients: This phase 2 study involved 33 women with metastatic or surgically incurable advanced breast cancer. They were all post-menopausal. The median age was 61 years, ranging from 39 to 77.

Treatment: Patients were treated with tipifarnib and fulvestrant. Tipifarnib is a small molecule that inhibits other molecules in cancer cells from being active. Fulvestrant is a drug that targets the estrogen receptor.

Toxicity: Five grade 4 toxicities were reported; two reports of neutropenia and one each of thrombosis, myocarditis, and pericardial effusion. Other toxicities (grade 1-3) included anemia, infection, nausea/vomiting, anorexia, fatigue, insomnia, neuropathy, and dyspnea.

Results: The median overall survival was 19.4 months.
