Patients: This Phase I study involved a total of 20 patients with newly diagnosed glioblastoma multiforme.

Treatment: Treatment consisted of administration of the chemotherapy drug, topotecan (hycamtin) with three-dimensional (3d) conformal radiotherapy.

Toxicity: Toxicities included neutropenia (grade 1-4), anemia (grade 1-3), and thrombocytopenia (grade 1-4). Other toxicities included febrile sinusitis, bacterial sepsis, nausea, vomiting, weight loss. (Two cases of pulmonary embolism were documented, one of which was fatal. However, the study “elected not to regard [this event] as dose limiting toxicity. Thromboembolic disease is rather common among patients suffering from malignancies and receiving chemotherapy in general.”)

Results: Overall survival was not discussed.
