Description:

Patients: This Phase II study involved 19 patients with metastatic colorectal cancer. 17 had colon cancer and 2 had rectal cancer. Nine patients were men, ten patients were women, and the median age was 62. All had received chemotherapy prior to the study.

Treatment: The treatment consisted of one chemotherapeutic agent: bortezomib.

Toxicity: No grade 4-5 toxicities were observed. Grade 3 toxicities included lymphopenia, abdominal pain, vomiting, and sensory neuropathy.

Results: The median overall survival was 28.3 weeks (6.5 months). No objective responses were observed, and three patients had stable disease. The authors concluded,"bortezomib... does not have clinical activity in patients with metastatic colorectal cancer and should not be investigated further in clinical trials as a single agent."
