Description:

Patients: This Phase III trial involved 179 patients with metastatic breast cancer whose cancers did not grow or spread (progress) after being treated with chemotherapy.

Treatment: Of the 179 patients, 114 patients were administered oral marimastat. Marimastat is an agent that inhibits matrix metalloproteinases (MMP’s). MMP’s are believed to facilitate tumor growth.

Toxicity: One patient who received marimastat died of presumed intracerebral hemorrhage. (This patient was known to have a large cerebellar metastasis.) In addition, a patient who received placebo died of a pulmonary metastasis. These deaths and others were not attributed to the drug. Grade 3 toxicities included pain and inflamed joints and tendons.

Results: Overall median survival was better in the placebo group (26.6 months) than in the marimastat group (24.7 months).
