Description:

Patients: This phase 2 study involved 151 people with locally advanced non-small cell lung cancer. There were 78 men and 73 women. The median age was 62, ranging from 43 to 78 years.

Treatment: Patients were treated with chemotherapy and radiation, on different schedules. Patients in Arm A were treated with paclitaxel and carboplatin then accelerated radiation therapy and more paclitaxel and carboplatin. Those in arm B were treated with paclitaxel and carboplatin then daily paclitaxel in combination with radiation therapy. Patients in Arm C were treated with paclitaxel and carboplatin then weekly paclitaxel and radiation therapy.

Toxicity: Grade 3 and 4 esophagitis and pneumonitis were reported. Grade 3 and 4 esophagitis was reported in 18 patients, and pneumonitis in two. Other, less severe toxicities were hypersensitivity, neurotoxicity, myalgia, nausea, and alopecia.

Results: Median survival was 17.8 months (17.7, 17.7 and 20.6 months for A, B and C, respectively).

Support: This study was supported by Bristol-Myers Squibb, which markets paclitaxel as Taxol and carboplatin as Paraplatin.
