Description:

Patients:
This phase 2 study involved advanced malignant pleural mesothelioma patients who had not received prior chemotherapy and who were divided into two separate treatment groups. Group A consisted of 40 patients with a median age of 64 years, 80% of whom were male. Group B had 23 patients with a median age of 66 years, 87% of whom were male.

Treatment:
Patients in group A received chemotherapy with pemetrexed and cisplatin, and with CBP501, a biologic therapy agent that interacts with a protein called calmodulin to allow more of the chemotherapy drug into the tumor cells.

Patients in group B received chemotherapy with pemetrexed and cisplatin only.

Toxicities:
There was one treatment-related death in group A due to respiratory difficulties. Grade 3-4 fatigue and vomiting, and grade 1-2 infusion-related reactions were also reported.

The most severe toxicities in group B were of grade 4 and included grade 3-4 fatigue, dehydration, and abdominal pain.

Results:
The median overall survival rates for groups A and B were 13.3 and 12.8 months, respectively.
