Description:

Patients: This phase II study involved 55 women with metastatic breast cancer. They ranged in age from 37 to 76 years. They were divided into two groups; those who had previous treatment with chemotherapy as an adjuvant and those who received chemotherapy for metastatic disease.

Treatment: All patients were treated with the chemotherapy drug 10-EDAM. It is chemically similar to methotrexate.

Toxicity: Grade 3/4 granulocytopenia occurred in 25% of patients, and stomatitis in 20%. Other grade 3/4 toxicities (occurring less frequently) were anemia, anorexia, fatigue, leukopenia, thrombocytopenia and increased levels of transaminase.

Results: The median overall survival for all patients was 11.8 months. For women in group 1 (women who received chemotherapy before as an adjuvant), all patients died and the median overall survival was 12.8 months (ranging from 2.5 to 64.5 months). In group 2 (women who received previous chemotherapy for metastatic disease), the median overall survival was 11.6 months (ranging from 3.8-69.5), and 33 of the 34 women have died.
